Biography

I am a basic cellular and molecular immunologist whose approach to immunology has been heavily influenced by experimental hematology, that is that lymphocytes are the progeny of hematopoietic stem cells and that their development follows many of the general rules of hematology. For my PhD, carried out at the Walter and Eliza Hall Institute, Australia in the group of Prof. Don Metcalf, I characterized the ontogeny of so-called "alloreactive" T cell precursors in the developing mouse thymus. Alloreactive T cells are involved in allogeneic graft rejection. Part of my work at this time (1979) was also to show that non-cytotoxic (CD8-) T cells were responsible for allogeneic mouse skin graft rejection. For my post-doctoral studies at the Ludwig Institute for Cancer Resaerch, Lausanne, Switzerland I was one of the first to combine flow cytometry, monoclonal antibodies and limiting dilution to quantify the alloreactive T cell response. We also identified for the first time, early T cell progenitors in the thymus. In Canberra, Australia I was responsible for establishing a functional flow cytometry facility at the John Curtin School of Medical Research. With Peter Doherty, we dissected the inflammatory infiltrate in mouse lymphocytic choriomeningitis virus infection and showed that this immune response in vivo obeyed the now-accepted rules of MHC restriction. In collaboration with Charmaine Simeonovic, we described immunological aspects of tolerance induction to transplanted xenogeneic (pig) pro-islets. In the LGME institute directed by Pierre Chambon in Strasbourg, France I learnt how molecular biology could impact on my own research interests. There, we generated a transgenic mouse over-expressing the cytokine Interleukin-7 (IL-7), a molecule involved both in lymphocyte development and survival. From these transgenic mice and in order to biologically clone cell lines from lymphomas that arose in these mice, we developed a bone marrow-derived mesenchymal stromal cell (MSC) clone. Since that time (1993), I have focused my research interest on IL-7. The advent of a recent mesenchymal stromal cell-based co-culture system supplemented with IL-7 for growing and influencing the commitment and development of lymphoid progenitors has revolutionized our approach to lymphocyte development. From 2003-2008, I was a guest scientist in the group of Prof. Ton Rolink at the University of Basel. Here, I collaborated with scientists investigating the immunological properties of MSC in vitro, work resulting in two publications. With Prof. Rolink, we were interested in the role of regulatory T cells in preventing auto-immunity that arises following bone marrow transplantation. Recently, with colleagues in Birmingham and Basel, I have become interested in more general aspects of lymphocyte development and hematopoietic lineage commitment. This has resulted in several publications, a book and recently attribution of an FP7 Marie Curie Initial Training Network grant entitled "DECIDE" with which we hope to further develop our modifiied model of hematopoiesis.
In addition to science, I'm interested in Irish traditional music. I play the fiddle. I'm also interested in hill walking and fitness.

Research Interests
