Intra-articular cartilage regeneration using stem cells loaded composite scaffolds.

Cartilage has a limited ability to repair after injury and this lack may ultimately lead to the development of osteoarthritis in affected joints. However, successful intervention after traumatic injury to joint cartilage may prevent or slow the onset of osteoarthritis. To date tissue engineering of cartilage has met with little success because of limited mechanical integrity and abnormal expression of cartilage collagens in the repair tissue. This project will test the hypothesis that articular hyaline cartilage can be restored using scaffolds seeded with 1) stem cells specifically mesenchymal stem cells (MSC) or 2) gene vectors or stem cells transduced to express genes that either promote hyaline cartilage formation or prevent formation of inappropriate collagens. We will work with traditional scaffolds or novel nano-scaffold materials to treat chondral or osteochondral defects which will be created in a rabbit joint as a model of human disease. The aim of the project will be to reverse the articular damage and create the potential to change the way we treat all articular diseases in humans.
