Research Interests

The overarching research theme of the FitzGerald group is the investigation of the pathological processes occurring during multiple sclerosis. The discovery of what is going wrong, both in the central nervous system and the rest of the body, is key to understanding the disease and can lead us towards the following research ideals:
1. being able to develop therapies for the treatment of MS
2. aiding in the speed and accuracy of disease diagnosis
3. improving the quality of life of the person with MS.

Of particular interest to the group is a cellular response known as endoplasmic reticulum stress (ER stress). This stress occurs when the cell is overloaded (e.g. during the production of myelin) and also under pathological conditions, such as those occurring during the development of MS, Parkinson’s disease, Alzheimer’s disease and following brain ischaemia (stroke). The ER stress response initiates a cascade of events called the unfolded protein response which aims to restore balance within the cell. Under chronic activation, however, cell death can occur.  Dr. FitzGerald's group has spent many years investigating the role of ER stress in the pathology of white matter and grey matter lesions both in human post-mortem tissue and in an animal model of MS. The group has also examined the role of ER stress in the process of myelin production, something that can occur inefficiently or aberrantly in MS following a demyelination event. Similarly, ER stress responses to iron overload in brain (a potential contributor to MS pathology and other neurodegenerative disorders) have been examined using brain slice-culture technology.

In an effort to expand translational research activities, body fluid samples obtained from individuals with MS, other neurological disorders, non-inflammatory disorders and from healthy individuals, are being analysed for the presence of proteins that could represent novel disease biomarkers. This is a long-term project that requires the collection, storage and testing of a large number of samples. The group is also exploring the possibility of collecting and storying skin punch biopsy samples from pwMS, with a view to producing induced pluripotent stem cells and brain cells that could be useful as a treatment management tool.

In an EU-funded project, the group has recently joined a consortium of early-stage researchers and principal investigators, focused on the development of novel biomaterials-based therapies for Parkinson's disease.

The above projects are being carried out in collaboration with researchers based at NUIG, in Imperial College London, the University of Innsbruck, the University of Glasgow, Monash University and with clinicians based at University Hospital Galway, and St. Vincent's hospital, Dublin.
